The viscoelastic properties of isolated segments of rabbit aorta were investigated using dynamic and equilibrium stress relaxation tests. Previous studies have shown that mechanical stability of aortic segments increases in cholesterol-induced hypercholesterolemia. This study addresses how the elasticity of the aorta is affected by cholesterol. Aortic segments from rabbits fed 1% cholesterol for 6 weeks were compared with segments from rabbits fed the cholesterol diet for 3 weeks, which had cholesterol-cholesterol ester levels equivalent to segments from cholesterol-fed animals, but triglyceride levels equivalent to those in the 3-week control rabbits. The stress relaxation responses of the aorta were measured on segments of 60% thoracic aorta mounted in a perfusion rig using a custom-built pressure transducer-tube system. A pulse-waveform was superimposed on the continuous data at peak stress. The main features of the stress relaxation responses in segments from cholesterol-fed rabbits were similar to those in controls; at high stress levels the responses were slightly slower. In the cholesterol-fed rabbits, the aortic segments were stiffer than in controls. The elastic modulus was slightly lower for segments from cholesterol-fed rabbits when the intact segments were loaded at lower stress levels but there were no significant differences for this condition when the segments were loaded at higher stresses. It is concluded that the increased stiffness in cholesterol-fed rabbits is not due to the cholesterol content of the aorta, but may be due to the abnormal changes in
